BREAKWATER Study Highlights the Growing Impact of Personalised Medicine in Colorectal Cancer

At this year’s ASCO Annual Meeting, researchers presented new results from the Phase III BREAKWATER trial, providing further evidence that a personalised treatment approach can improve outcomes for people with BRAF V600E-mutated metastatic colorectal cancer (mCRC).

BRAF V600E is a genetic alteration found in approximately 8–10% of patients with mCRC and is associated with a more aggressive disease course and poorer outcomes compared with other forms of colorectal cancer.

The latest BREAKWATER data evaluated a treatment combination consisting of encorafenib, cetuximab, and FOLFIRI chemotherapy in the first-line setting. Results showed a significant improvement in progression-free survival compared with standard treatment, while also demonstrating higher response rates.

These findings build on previous BREAKWATER results that supported the use of targeted therapy combinations in patients with BRAF V600E-mutated mCRC and further strengthen the role of biomarker-driven treatment strategies.

For patients, the message is clear: knowing the molecular profile of a tumour can make a real difference to treatment options and outcomes.

Today, testing for biomarkers such as BRAF, RAS and MSI status is an essential part of colorectal cancer care, especially in advanced stages. These tests help identify patients who may benefit from targeted therapies or immunotherapy and support more personalised treatment decisions.

While the new BREAKWATER results are primarily relevant to a specific subgroup of patients, they reflect a broader shift in colorectal cancer care from a one-size-fits-all approach towards treatments tailored to the biology of each person’s tumour.
